Assessing the Photo-degradation of Biopharmaceutical Compounds by UV-Vis (CAT#: STEM-B-0396-CJ)

Introduction

All biopharmaceutical compounds are sensitive towards chemical degradation. Examples are deamidation, hydrolysis, oxidation, photo-degradation, disulfide-scrambling, and others. Chemical degradation can lead to aggregation, charge variants and/or structural changes of the drug substance and eventually impair the effectivity or safety of the therapy.




Principle

The Principle of UV-Visible Spectroscopy is based on the absorption of ultraviolet light or visible light by chemical compounds, which results in the production of distinct spectra. Spectroscopy is based on the interaction between light and matter.

Procedure

1. Sample preparation.
2. Pass UV-visible light through the sample to measure the transmittance of the sample to light.
3. From transmittance (T), absorbance can be calculated as A=-log (T). An absorbance spectrum shows the absorbance of a compound at different wavelengths.
