Ion-Exchange Chromatography Technology

Ion-Exchange Chromatography Technology is a chromatographic method in which ion-exchange resin or chemically bonded ion exchanger is used as the stationary phase and the difference of ion exchangeability or total charge of the separated components is used to achieve separation. This technique enables the separation of similar types of molecules that would be difficult to separate by other techniques because the charge carried by the molecule of interest can be readily manipulated by changing buffer pH.

Procedure
  • Packing of chromatographic columns
    The ion exchange separation process is usually carried out in chromatographic columns packed with an ion exchanger.
  • Loading of the sample
    After the column is packed, the solution to be assayed is placed on top of the column and allowed to pass through the bed of the ion exchanger with the help of a syringe or pipette.
  • Developments of the chromatogram
    Chromatograms were developed with the aid of different mobile phases.
  • Analysis of detection of the eluate
    Collecting the different components of the eluate and analyzing its content.
  • Regeneration
    After separation, it should not be disposed of due to its cost.
