Surface Plasmon Resonance (SPR) is a powerful optical phenomenon that occurs when polarized light, under total internal reflection, excites free electrons at the surface of a thin metal film. Any change in the refractive index near the metal surface alters the resonance angle (SPR angle) of the incident light. By continuously recording these changes, detectors can capture the dynamic process of biomolecular interactions in real time within microfluidic channels attached to the chip surface.

Phase Meaning
Baseline The initial signal when buffer flows across the sensor surface before testing begins.
Association The association process as analytes bind to immobilized ligands.
Equilibrium The point where binding and dissociation reach dynamic balance.
Dissociation The process of buffer washing away bound analytes.
Regeneration Flushing the chip surface to remove remaining analytes while preserving ligand activity for subsequent runs.
