Analysis of Proline (Pro) by Ultraviolet-visible Spectrophotometer (CAT#: STEM-MB-1327-LGZ)

Introduction

Sensitivity: 0.17 μg/mL
Detection range: 0.17-35 μg/mL
Precision: inter-lot difference of 5.4%, intra-lot difference of 4.5%
Detection equipment: Ultraviolet-visible Spectrophotometer
Detection wavelength: 520 nm

Principle

Only proline (Pro), an amino acid in plants, reacts with acidic ninhydrin to produce a stable red product. The product has a maximum absorption peak at 520 nm wavelength, and the absorption value is linear with the content of proline. Therefore, the content of proline in samples can be determined by acid ninhydrin method.

Applications

Used to detect the content of proline in plant tissue and honey samples.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.

Materials

• Sample Type: Plant tissue, honey sample
