The progression of pancreatic cancer (PC) is significantly associated with tumor immune escape, possibly linked to dysfunction in natural killer (NK) cell activity. CD226, CD96, and TIGIT share the ligand CD155 and play crucial roles in regulating NK cell function. This study aims to explore the roles of these molecules in NK cells of PC patients. TIGIT and CD96, along with the co-stimulatory receptor CD226, form a pathway resembling the CD28/CTLA-4 pathway, sharing ligands and distinct receptors. The ligand affinity in this pathway finely tunes immune responses. Although the roles of TIGIT and CD96 as immune checkpoint receptors in T cells and NK cells are just beginning to be understood, accumulating data support their targeted use to enhance anti-tumor immune responses. A clear understanding of the immune cell populations regulated by TIGIT and CD96 is crucial for the design of immunotherapies that combine these receptors with existing immune checkpoint blockade therapies. Dysfunction of CD96 may lead to C syndrome, a rare syndrome characterized by multisystem defects, including trigonocephaly, developmental delay, cardiac defects, and distinctive facial features.
