Hybridoma Antibody Gene Sequencing (CAT#: STEM-MB-0126-WXH)

Introduction

Hybridoma cells are prone to problems such as contamination, gene loss, poor cell status and even death during storage, which will affect the quality of the antibodies produced. By extracting and sequencing the antibody genes in the hybridoma cells, the corresponding nucleotide sequences are obtained to facilitate the stable expression and production of exogenous antibodies in the later stage.

• Permanently retain monoclonal antibody sequence information to prevent hybridoma cells from losing monoclonal antibody information.
• Helps the selection of target cell lines and expresses recombinant monoclonal antibodies with higher purity.
• Can shorten the time-consuming process of hybridoma cell cloning and speed up the production of monoclonal antibodies.

Procedure

1. Hybridoma Single Cell Array
2. RT-PCR to obtain cDNA
3. Amplify antibody light and heavy chain genes (and exclude light chain pseudogenes)
4. Link the resulting gene to the vector
5. Sequencing
6. Bioinformatics analysis
