Principle

Superoxide anion radical (O2.-) is produced through the xanthine and xanthine oxidase reaction system, which oxidizes hydroxylamine to form nitrite and produces purplish red compounds under the action of chromogenic agents. When SOD was added into the reaction system, the disproportionation of superoxide anion radical was catalyzed, and the formation of nitrite was reduced, and then the purplish red was lighter. SOD activity was calculated according to the absorbance difference between the sample tube and the charge. Meanwhile, Mn-SOD was inactivated in samples treated with the reagent, but CuZn-SOD was not affected.

Applications

It is suitable for detecting the activity of T-SOD, CuZn-SOD and Mn-SOD in serum (plasma), urine, cells and cell supernatant, various animal and plant tissues.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.
