This immobilized-metal affinity medium is designed for purification of His-tagged recombinant proteins and compatible metal-binding molecules. It uses a polymethyl methacrylate matrix with a hydrophilic surface modification. IDA chelating groups immobilize nickel ions on the particle surface. Histidine-containing proteins bind selectively to the coordinated nickel during chromatography. The medium can also process compatible peptides, nucleotides, and phosphorylated proteins. Its 1000 Å pore structure supports transport of biological macromolecules into the particles. The rigid 80 µm particles provide high mechanical stability and recommended linear flow up to 800 cm/h. The stated His-protein capacity of at least 25 mg/mL supports preparative affinity purification.
