This affinity chromatography medium is designed for selective capture of messenger RNA from complex mixtures. It uses a highly crosslinked polystyrene-divinylbenzene matrix with a hydrophilic surface treatment. Covalently attached Oligo dT(20) groups bind specifically to Poly(A) tails through base pairing. The hydrophilic surface minimizes nonspecific interactions with other sample components. Uniform 50 µm particles support consistent packed-bed performance and rapid mass transfer. The rigid matrix provides high salt tolerance, pressure resistance, and flow above 400 cm/h. Captured mRNA can be released under standard purification conditions. The medium is chemically stable in concentrated salt, selected alcohols, acetonitrile, and isopropanol solutions.
