In Vitro Models of Angiogenesis (CAT#: STEM-CBT-0032-WXH)

Introduction

In vitro models of angiogenesis can be divided into proliferation experiments, migration experiments, and tube formation experiments at the cell level, as well as human placental vessel segment culture models and rat arterial ring models at the organ level. At present, the angiogenesis experiment commonly referred to refers to the lumen formation experiment. Lumen formation reflects the early process of capillaries and is the most complete indicator of endothelial cell function in vitro.<br />Drugs inhibit the formation of blood vessels by inhibiting the formation of the lumen or disrupting the formed lumen. With the help of computer software, the number of small tubes, the number of connections between small tubes, the length and area of small tubes can be calculated, and the influence of drugs on the formation of the lumen can be quantitatively analyzed.

Procedure

1. Matrigel Preparation
2. Collection of Cells
3. After the gel is solidified, add the cell suspension to the well plate
4. Routine cell culture
5. Analysis of results: 5 visual fields were randomly selected under a 100X microscope to observe and count the number of vascular endothelial cell tubes. Tube formation rate = the number of experimental tubes / the number of tubes in the control group * 100%
