Description

Glutathione-superflow allows rapid affinity purification of GST-tagged proteins for batch, gravity flow, and FPLC applications. The resin is based on 6% cross-linked agarose, with glutathione covalently bound to the resin. It can withstand flow rates and media pressures up to 15 ml/cm2/min (up to 150 psi).

The GST purification kit contains sufficient buffer and glutathione-superflow columns to perform five batches/gravity flow purifications. Up to 10 mg of gst-tagged recombinant protein can be purified using this kit.

GST tags

Epitope tags, such as glutathione-S-transferase (GST), are commonly used as tags for protein expression and purification applications. Glutathione transferase, an abundant enzyme involved in cellular defense against electrophilic compounds, binds glutathione with high affinity and specificity. The strength and selectivity of this interaction can be exploited for efficient purification of GST-tagged proteins. Glutathione-Superflow resin selectively binds GST-tagged proteins under normal conditions, allowing rapid and efficient isolation of proteins of interest from whole-cell extracts. High purity is achieved in a single chromatographic step.

GST is a 35kda protein that can be detected by biochemical and immunological methods. This feature sets it apart from many epitope tags, which are simply short peptides. However, since GST is a large tag, the potential for protease degradation is higher than for small tags. Therefore, it is imperative to perform GST-based protein purification as quickly as possible under non-degrading conditions to minimize sample loss. GST loses its ability to bind to glutathione-superfluid resin when it is denatured, so strong denaturants such as guanidine-hydrochloric acid or urea cannot be used in the purification buffer. When designing a purification protocol, check reagent compatibility.
