Cellular ROS Detection (CAT#: STEM-CBT-0003-WXH)

Introduction

Reactive oxygen species are free radical active substances produced in mitochondrial oxidative respiration of cells, and participate in various physiological and pathological mechanisms of cells. Detecting the content of ROS in cells can understand the changes of ROS in some physiological and pathological states of cells, and analyze drug effects and related mechanisms.<br />The detection of reactive oxygen species was performed using the fluorescent probe DCFH-DA. Active oxygen in cells can oxidize non-fluorescent DCFH to produce fluorescent DCF, and the level of active oxygen in cells can be known by detecting the fluorescence of DCF.

Procedure

1. Make a single cell suspension (tissue sample)
2. Hemolysis (this step is not required for cell samples)
3. Centrifuge to remove supernatant
4. Adjusting Cell Density
5. Add the corresponding fluorescent probe
6. Incubation
7. After washing, centrifuge to remove the supernatant
8. On-machine detection (flow cytometer, microplate reader)
