xTractor Buffer is the most flexible and efficient cell lysis buffer for protein extraction from bacterial, yeast, mammalian, and baculovirus-infected cells. This cell lysis reagent provides rapid and efficient (10 minutes) lysis using mild, non-denaturing conditions. xTractor Buffer has been optimized for extraction of HE-tagged proteins, so it is compatible with all IMAC resins.

Cell lysis is simple. Simply resuspend the cell pellet in cell lysis buffer (1:20 w/v) and mix gently for 10 minutes, reducing the risk of proteolysis. The final lysate has no visible cell fragments or pellets. After extraction, simply centrifuge or filter the lysate and load it on any affinity resin column to isolate the labeled protein. To save time, the resulting lysate can be loaded directly onto a TALON CellThru column without centrifugation or filtration.

This cell lysis protocol is an efficient method to extract protein from any cell for higher protein purity and yield. Compared with sonication, xTractor Buffer extraction yields more protein content from the same size sample. In addition, the biological activity of the protein extracted by xTractor Buffer is higher than that of the protein obtained by sonication.

The xTractor Buffer Kit provides enough buffer to extract protein from 10 grams of bacterial pellet. The buffer kit also includes lysozyme and DNase I. This method is especially useful for high molecular weight proteins such as β-galactosidase (LacZ), which cannot be extracted unless the membrane is completely disrupted.

xTractor Buffer has been optimized for extraction of tagged proteins, which can then be purified with any of our affinity resins, such as TALON Co resin or His60 Ni Superflow resin, or any other affinity resin.
