The Protein Thiol Immobilization Kit is a complete kit designed for the simple and efficient coupling of proteins to solid agarose supports. Thiol-coupled resin columns utilize iodoacetyl groups, which specifically react with free thiols to form covalent, permanent thioether linkages. Long spacer arms reduce steric hindrance, ensuring greater binding of protein and antibody during affinity purification.

Proteins, including antibodies, must have free sulfhydryl groups in order to be immobilized on the resin. A mild reducing agent, 2-mercaptoethylamine, is used to reduce the disulfide bonds in the hinge region of antibodies while preserving the functionally critical disulfide bonds between the heavy and light chains.

The resulting columns can be used to study protein-protein interactions or for purification by affinity chromatography. Depending on the stability of the immobilized molecules, the column can be used multiple times without significant loss of activity.
