This Protein A affinity medium is designed for capture and purification of monoclonal and polyclonal antibodies. It uses highly crosslinked spherical agarose microspheres as the base matrix. An alkali-resistant recombinant Protein A ligand is immobilized on the matrix. The ligand selectively binds the Fc region of antibodies. One affinity step can recover high-purity antibodies from ascites, serum, or cell-culture fluid. A stated binding capacity of 70 mg/mL supports productive capture operations. The medium provides flow above 250 cm/h under the specified pressure condition. Resistance to sodium hydroxide supports repeated cleaning and reuse. Comparative testing showed antibody purity, recovery, and capacity close to the referenced benchmark medium.
