This Protein A affinity medium is designed for large-scale antibody manufacturing. It uses a rigid polymethyl methacrylate matrix with accurately controlled porosity. A long hydrophilic spacer arm reduces steric hindrance and nonspecific adsorption. Alkali-resistant recombinant Protein A selectively binds the Fc region of antibodies. The medium provides a dynamic human polyclonal IgG capacity of 55-65 mg/mL at five minutes residence time. Its 50 µm monodisperse particles support high resolution and stable packed-bed performance. The rigid matrix allows flow up to 800 cm/h and operating pressure up to 0.8 MPa. The medium is suitable for monoclonal and bispecific antibody capture with alkaline cleaning between cycles.
