An important tool in research is the generation of peptide and protein affinity columns for purification of antibodies and discovery of important interacting proteins and cofactors. G-Biosciences provides a complete kit for coupling peptides and proteins to agarose via their primary amines. These kits contain all the reagents to create a stable bond, resulting in a column that can be reused many times.

The Amine Reactive HOOK™ Agarose Coupling Kit is designed for simple and efficient coupling of proteins and peptides to solid supports. This kit allows the generation of stable and reusable columns for the affinity purification of antibodies, proteins and other protein-interacting biomolecules.

This kit utilizes our HOOK™ Activated Sepharose (amine), which has reactive aldehyde groups. The aldehyde groups of agarose react spontaneously with primary amines on the N-terminus of proteins or lysine residues to form intermediate Schiff base complexes. These, in turn, are selectively reduced by the reducing agent provided. After the reaction is quenched, the protein or peptide is attached to the agarose via a stable amine linkage. The resin can now be mixed into the provided columns for purification and interaction studies.

In addition to all coupling reagents, HOOK™ Agarose Coupling Kit also provides a specific Optimizer Buffer™, which can provide optimal coupling conditions and can be used to equilibrate the initial protein sample.

This kit is suitable for the preparation of 5 stable columns containing 2ml resin.
