Con A agarose is made by coupling Con A and agarose by cyanogen bromide method. Con A is a tetrameric metalloprotein lectin isolated from Canavalia ensiformis (jack bean).

Con A is used in the purification of glycoproteins, polysaccharides and glycolipids as it binds molecules containing α-d-mannopyranyl, α-d-glucopyranyl and sterically related residues. Con A Agarose is also used in other applications including purification of enzyme-antibody conjugates, purification of IgM and isolation of membrane vesicles.

As mentioned above, Con A is a metalloprotein and the presence of Mn2+ and Ca2+ is essential in order to maintain its binding properties. Each subunit of Con A utilizes one calcium ion and one manganese ion, and these cations can be removed under acidic conditions, abolishing the carbohydrate binding activity.
