This high-performance hydrophobic interaction medium is designed for downstream purification of biological macromolecules. It uses small high-flow agarose microspheres functionalized with butyl ligands. The butyl groups provide hydrophobic selectivity under controlled salt conditions. Biomolecules bind according to exposed hydrophobic regions and elute as ionic strength decreases. The approximately 40 µm particles improve mass transfer and separation resolution. A dynamic HSA capacity of approximately 37 mg/mL supports productive preparative operation. The medium operates above 220 cm/h at the stated pressure and bed height. Broad pH stability supports repeated cleaning-in-place. It tolerates the listed alkaline solutions, alcohols, detergents, denaturants, and ammonium sulfate.
