Analysis of KISS1 Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-1108-LGZ)

Introduction

Official Full Name: KiSS-1 metastasis suppressor
Also known as: HH13; KiSS-1
This gene is a metastasis suppressor gene that suppresses the metastasis of melanoma and breast cancer without affecting tumorigenicity. The encoded protein can inhibit the chemotaxis and invasion of malignant melanoma, thereby attenuating tumor metastasis. Studies have shown that cytoskeletal reorganization may be involved in the regulation of events downstream of cell-matrix adhesion. The gene's protein product, kisspeptin, stimulates gonadotropin-releasing hormone (GnRH)-induced gonadotropin secretion and regulates pubertal activation of GnRH neurons. Polymorphisms in the terminal exons of this mRNA result in two protein isoforms. Adenosine at the polymorphic site represents the third position of the stop codon. When adenosine is missing, a downstream stop codon is utilized and the encoded protein is extended to an additional 7 amino acid residues.

Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Applications

Gene mutation analysis.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
