Introduction

Alpha-2-macroglobulin (A2M) is a proteinase inhibitor. A2M acts as a broad spectrum proteinase binding protein. It is a large plasma glycoprotein and has long been recognized as an irreversible inhibitor of many proteases. Recently, many growth factors, cytokines, and hormones have been reported to bind to α2M through a variety of mechanisms. A2M is also produced in the brain, where it binds to a variety of extracellular ligands and is internalized by neurons and astrocytes. In the brains of patients with Alzheimer's disease (AD), A2M is localized to diffuse amyloid plaques. A2M also binds to soluble beta-amyloid protein, where it mediates degradation. Clinical cirrhosis, nephrotic syndrome, diabetes, severe burn cases increased.
