Analysis of Methionine by Amino Acid Analyzer (CAT#: STEM-MT-2514-LGZ)

Introduction

Amino acids are the basic units of biological macromolecules protein and active peptides, and are also necessary substances to maintain normal life activities of organisms. In addition, the composition and content of amino acids also affect the efficacy of some medicinal substances and the taste and nutritional value of edible products. Amino acid composition analysis has been widely used in the study of amino acid composition and content of various edible and medicinal substances.




Principle

Taking advantage of the different structure, acidity, alkalinity, polarity and molecular size of various amino acid components in the sample, they are separated on a cation exchange column, and the amino acid components are eluted sequentially by using buffer solutions with different pH values and ion concentrations. Then mix them one by one with ninhydrin reagent from another flow path, and then flow them together into the spiral reaction tube, and carry out color reaction at a certain temperature (usually 115-120°C), forming a blue-purple product with maximum absorption at 570nm. Among them, hydroxyproline reacts with ninhydrin to form a yellow product, and its maximum absorption is at 440nm.

Procedure

1. Sample handling:
Determining the content of various free amino acids in the sample can be directly loaded on the column for analysis after removing fat impurities.
When determining the amino acid composition of protein, the sample must be hydrolyzed with acid to make the protein completely converted into amino acid before being loaded on the column for analysis.
2. Sample analysis:
The processed samples were loaded onto the column for analysis.
3. Result calculation:
We can calculate the exact amount of amino acids. The type of amino acid can be determined according to the time when the peak appears.
