Description

Syngene's GeneFlash is a relative newcomer at the US market of entry level CCD-based gel documentation systems. It is built around an 8-bit 768 X 582 pixel black-and-white camera with a microprocessor-controlled dark room. The latter accepts most small-to-medium bench-top UV transilluminators (up to 25 X 30 cm gels), or one can be supplied by Syngene. The large sliding door in the darkroom allows for easy access to the transilluminator. The dark room has an integrated color LCD display and an 8-key panel that allows you to manipulate the camera, printer, save files, and change system settings. People like me, who are getting tired of having to reach around and feel their way toward the On-Off buttons on the back panels of scientific instruments will welcome the Syngene designers' decision to put the 3 power switches (one main and two separate for the UV and white lights, respectively) on the front of the dark room enclosure. As a stand-alone unit, the GeneFlash can be used with a thermal video printer (one made by Sony is supplied by Syngene); alternatively it can be connected through ethernet or WiFi networks to a PC running GeneFlash server software. Gel pictures can be saved as BMP or TIFF files on a flash memory stick (USB port positioned next to the LCD) in a stand-alone mode or on the server when on the network. In addition to the server software, Syngene supplies a GeneTools package allowing you to perform adequate image analysis, background subtraction, etc. as well as some gimmicks (like re-colorization of the black-and-white images to appear in color for ethidium bromide- or SYBR® Green-stained gels). The CCD camera features manual focus, manual optical-only 8-48mm zoom, and manual aperture. The GeneFlash is supplied with a filter appropriate for ethidium bromide-stained DNA gels, but the optional filters available from Syngene are compatible with a wide variety of protein and DNA stains (Coomassie Blue, Silver Stain, SYBR® Gold, SYBR® Green, SYBR® Safe, GelStar®, Sypro® Red , Sypro® Ruby, Sypro® Orange, Fluorescein, Rhodamine Red™, Texas Red®, Pro-Q® Diamond , Deep Purple™). Non-fluorescent stains such as Coomassie require use of a dual (UV-Vis) transilluminator or UV-to-Vis converter.
In our lab, we are using GeneFlash system with a PULNiX TM-300 CCD camera and Computar H6Z0812 8-48 mm optics as well as many available options: Sony video graphic printer UP-895MD, built-in Ethernet for networking and USB port for flash memory. Most of the time the system is used for visualization and documentation of DNA in agarose gels: Molecular cloning and screening, PCR optimization, genomic DNA purification, preparation of templates for in vitro transcription, etc. The power-up time for the system is no more than 10 sec; it is easy to use the camera and navigate the menu. Most often a picture of a gel is printed out (quality is quite good); when needed, a digital copy is retrieved using a USB memory key or saved on the server for analysis with GeneTools software.
