Total Gene Synthesis (CAT#: STEM-MB-0079-WXH)

Introduction

Total gene synthesis refers to the technique of artificially synthesizing double-stranded DNA molecules in vitro. Gene synthesis is to design and synthesize overlapping single-stranded oligonucleotides according to the gene sequence of a certain protein, and then splicing the full length by overlapping extension PCR method. Gene synthesis does not require a template and is one of the means to obtain genes. Its advantages include:
1. The synthesis cycle is short and the accuracy rate is high
2. Greater flexibility
3. Genes that are difficult to obtain or even do not exist in nature can be designed
4. Codon optimization can be carried out so that the gene can be well expressed in various biological expression systems

Applications

• Cloning genes for which templates are not readily available
• Synthesizing new genes that do not exist in nature
• Heterologous gene expression
• Make primers for PCR technique

Procedure

1. Analysis, design and synthesis scheme
2. Synthesis of single-stranded oligo DNA
3. Splicing DNA Fragments
4. Cloning the full-length gene into the vector
5. DNA sequencing to confirm the correctness
6. Repair and Proofreading

Notes

• Customers provide the sequence of the gene to be synthesized.
• Customers explain the specific requirements of the experiment.
