Introduction

RNA Immunoprecipitation(RIP) is a technique for studying the combination of RNA and protein in cells, and it is a powerful tool for understanding the post-transcriptional regulatory network. This technique uses antibodies against the target protein to precipitate the corresponding RNA-protein complex, and then the RNA bound to the complex can be analyzed after separation and purification. RIP-Seq combines RIP technology with high-throughput sequencing, enabling high-throughput detection of RNA bound to specific proteins, thereby analyzing the target protein-RNA interaction network within the whole transcriptome.

Procedure

1. Collect cells
2. Isolate nuclei from nuclear pellets and lyse them
3. chromatin shear
4. Immunoprecipitate the target RNA binding protein (RBP) along with the bound RNA.
5. Purify RNA bound to immunoprecipitated RBP after washing off unbound material.
6. Convert RNA to cDNA and analyze using qPCR, microarrays, or sequencing.
