T-cell surface glycoprotein CD8 alpha chain, also known as CD8a, is a single-pass type I membrane protein. CD8 is expressed by thymocytes, mature T cells, and natural killer (NK) cells. It plays a crucial role in recognizing monomorphic determinants on major histocompatibility complex (MHC) Class I antigens and in signal transduction during T-cell activation. In T-cells, CD8 functions primarily as a coreceptor for the MHC class I molecule:peptide complex. These class I peptides are derived from cytosolic proteins. CD8 interacts simultaneously with the T-cell receptor (TCR) and the MHC class I proteins presented by antigen-presenting cells (APCs). This interaction recruits the Src kinase LCK to the vicinity of the TCR-CD3 complex, initiating various intracellular signaling pathways by phosphorylating different substrates. This process ultimately leads to the production of lymphokines, increased motility, adhesion, and the activation of cytotoxic T-lymphocytes (CTLs). This mechanism enables CTLs to recognize and eliminate infected cells and tumor cells. In NK cells, the presence of CD8A homodimers on the cell surface provides a survival mechanism that allows conjugation and lysis of multiple target cells. CD8A homodimer molecules also promote the survival and differentiation of activated lymphocytes into memory CD8 T-cells. Storage Information: Lyophilized samples can be stored at 2-8°C for 24 months. If aliquoting is necessary, the samples can be reconstituted with injection-grade water, physiological saline, culture medium, or PBS. After reconstitution, they should be aliquoted, stored at -20°C for 6 months, or at -80°C for 12 months. The post-dilution concentration should not be less than 100 μg/mL, and it is important to avoid repeated freeze-thaw cycles.
