This hydrophobic interaction medium is designed for process-scale purification of biological macromolecules. It uses high-flow agarose microspheres functionalized with phenyl ligands. The ligand provides strong hydrophobic interaction with exposed nonpolar regions on biomolecules. Binding is promoted under high-salt conditions and elution occurs as ionic strength decreases. The approximately 75 µm particles balance binding capacity and flow performance. Dynamic HSA capacity above 27 mg/mL supports productive downstream processing. The medium operates above 600 cm/h at the stated pressure and bed height. Broad pH and chemical stability support repeated regeneration and cleaning. Its scalable format is suitable for biopharmaceutical capture, intermediate purification, and polishing.
