HOOK™ 6X His Protein Purification Kit can purify soluble 6X His-tagged protein from yeast culture. Yeast is first lysed with Yeast PE LB™ and LongLife™ Zymolyase® to release total soluble protein while maintaining protein structure and activity. 6X His-tagged protein was purified by immobilized metal affinity chromatography (IMAC) by passing the clarified lysate through a pre-packed column.

Yeast-pe LB™ is used to extract soluble proteins from yeast cells. Yeast PE LB™ is a patented improvement to Zymolyase®-based spheroplast preparation and extraction of soluble proteins from yeast cells. Yeast PE LB™ is based on organic buffers utilizing mild non-ionic detergents and a proprietary combination of various salts and agents to enhance protein extraction and stability. A ready-to-use formulation of Zymolyase® is also provided. Depending on the application, other agents such as protease inhibitors can be added to Yeast PE LB™. This proprietary combination of reagents provides a simple and versatile method for yeast protein extraction. Yeast PE LB™ eliminates the need for glass bead lysis of yeast cells.

HOOK™ 6X His Protein Purification Kit can use nickel chelate immobilized metal affinity chromatography (IMAC) column or cobalt chelate IMAC column. Cobalt chelate resins have a lower binding affinity for 6X His tags compared to nickel chelate resins, which results in less non-specific binding and may result in slightly lower yields.

HOOK™ 6X His Protein Spin Purification Kit is optimized to obtain 10 mg of soluble His-tagged protein with a purity of 80-90%, depending on the protein's expression level, resin type, conformation, and solubility characteristics.
