HOOK™ 6X His Protein Purification Kit purifies soluble 6X His-tagged protein from bacterial culture. Bacteria are first lysed with bacterial PE LB™ and PE LB™-lysozyme, releasing total soluble protein while maintaining protein structure and activity. 6X His-tagged proteins were purified by immobilized metal affinity chromatography (IMAC) through pre-packed columns through clarified lysates.

Bacteria - PELB™ Kit has been developed for the extraction of soluble proteins from bacterial cells. It is based on a patented improvement of lysozyme lysis, allowing extraction of soluble proteins while removing nucleic acids (DNA and RNA) released during cell lysis. Bacteria-pe LB™ Lysis eliminates viscosity build-up, allowing efficient clarification with lower centrifugal forces.

HOOK™ 6X His Protein Purification Kit can use nickel chelate immobilized metal affinity chromatography (IMAC) column or cobalt chelate IMAC column. Cobalt chelate resins have a lower binding affinity for 6X His tags compared to nickel chelate resins, which results in less non-specific binding and may result in slightly lower yields.

HOOK™ 6X His Protein Purification Kit is optimized to produce 10mg/250ml of soluble His-tagged protein with a purity of 80-90%, depending on the protein's expression level, resin type, conformation and solubility characteristics.
