This heparin affinity medium is designed for purification of biological molecules that bind sulfated polysaccharides. It uses a crosslinked agarose matrix coupled with heparin. The ligand can interact with antithrombin III, coagulation factors, protein-synthesis factors, lipoproteins, interferons, and nucleic-acid-binding proteins. It is also applicable to restriction enzymes, thrombin, and thrombin-like proteins. Spherical particles provide uniform packing and convenient operation. The medium provides an antithrombin III adsorption capacity above 3 mg/mL. Broad chemical compatibility supports exposure to denaturants, alkaline solution, nonionic detergents, and glycerol. The matrix can withstand repeated heat treatment under the specified phosphate-buffer conditions.
