Caspase-3 Detection to analyze cell apoptosis (CAT#: STEM-CBT-0010-WXH)

Introduction

Caspase-3 mediates apoptosis through the extracellular pathway and the intracellular pathway, the extracellular pathway is called the death receptor pathway, and the intracellular pathway is called the mitochondrial pathway. The methods for detecting Caspase-3 activity in apoptosis experiments mainly include Western blot analysis, fluorescence spectrophotometer analysis, flow cytometry analysis, enzyme-linked immunoassay (ELISA) and so on.<br />Caspase-3 normally exists in the cytoplasm in the form of zymogen (32KDa), and it is activated in the early stage of apoptosis. The activated Caspase-3 consists of two large subunits (17KDa) and two small subunits ( 12KDa), cleave the corresponding cytoplasmic and nuclear substrates, and eventually lead to apoptosis. But in the late stage of apoptosis and dead cells, the activity of caspase-3 decreased significantly.
