Analysis of Alanine Aminotransferase (ALT/GPT) Activity by Fluorescence Microplate Reader (CAT#: STEM-MB-1508-LGZ)

Introduction

Sensitivity: 0.01 U/L
Detection range: 0.01-0.83 U/L
Precision: inter-lot difference of 9.800000000000001%, intra-lot difference of 2.3%
Detection equipment: Fluorescence Microplate Reader
Excitation wavelength: 535 nm
Emission wavelength: 587 nm

Principle

Alanine aminotransferase (ALT) can catalyze the transamination of alanine and α-ketoglutaric acid to produce pyruvate and glutamic acid. Under the action of pyruvate oxidase, pyruvate generates H2O2. Under the action of peroxidase, the non-fluorescent probe is oxidized into a fluorescent substance. The activity of ALT can be calculated by measuring the increase of the fluorescence value of the system.

Applications

It is suitable for the detection of alanine transaminase activity in animal tissue samples and serum (plasma) and other liquid samples.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Determination of fluorescence intensity.
4. Make the mark curve and calculate the result.

Materials

• Sample Type: animal tissue samples and serum (plasma) and other liquid samples
