Analysis Ligand-receptor Binding of Biopharmaceutical Compounds by Surface Plasmon Resonance (SPR) (CAT#: STEM-B-0395-CJ)

Introduction

Biological activity is, besides the safety of the therapy, the most important aspect of a drug product or therapy. Thus, a major goal of formulation development is to maintain the functionality of the therapeutic compound inside its formulation throughout its shelf-life.

Binding of a ligand to a receptor changes its shape or activity, allowing it to transmit a signal or directly produce a change inside of the cell. Stages of signal transduction: ligand-receptor binding, signal relay, response.

Principle

General Principle of SPR. Surface plasmon resonance occurs when a photon of incident light hits a metal surface (typically a gold surface). At a certain angle of incidence, a portion of the light energy couples through the metal coating with the electrons in the metal surface layer, which then move due to excitation.

Applications

Biopharmaceutica

Procedure

1. Detect Buffers and prepare samples.
2. Set up and run the experiment.
3. Collect and analyse data.

Materials

• Sample: Peptides, Proteins, Vaccines, Virus-like particles
• Equipment: Surface Plasmon Resonance (SPR)

Notes

Surface plasmons have been used to enhance the surface sensitivity of several spectroscopic measurements including fluorescence, Raman scattering, and second-harmonic generation. However, in their simplest form, SPR reflectivity measurements can be used to detect molecular adsorption, such as polymers, DNA or proteins, etc.
