Introduction

Lectin is a protein that can be synthesized and secreted by both animal cells and plant cells, and plays an important role in cell recognition and adhesion reactions, mainly promoting intercellular adhesion. Lectin has more than one sugar-binding site, so it can participate in cell recognition and adhesion, and connect different cells.




Applications

For the purification of lectins.

Procedure

1. Column packing: choose prepacked column or manual column packing.
2. Sample treatment: Hydrophobic chromatography is loaded under the condition of high salt ion strength. Moreover, it is necessary to pay attention to the fact that there is no sediment before the sample is loaded, and if there is any, it should be removed by centrifugation.
3. Column pre-equilibration: Wash the column with 10 times of chromatography buffer to balance it.
4. Loading the sample: close the liquid outlet, and then load the sample.
5. Washing: Wash off the impurity proteins that are not adsorbed on the column as much as possible.
6. Elution: Use a gradient mixer for gradient elution or configure a series of elution buffers with two volumes for each wash.
7. Collection of samples.
8. Check of sample purity.
